Metabolic protein launches sugar feast that nurtures brain tumors

Researchers at The University of Texas MD Anderson Cancer Center have tracked down a cancer-promoting protein's pathway into the cell nucleus and discovered how, once there, it fires up a glucose metabolism pathway on which brain tumors thrive.

They also found a vital spot along the protein's journey that can be attacked with a type of drug not yet deployed against glioblastoma multiforme, the most common and lethal form of brain cancer. Published online by Nature Cell Biology, the paper further illuminates the importance of pyruvate kinase M2 (PKM2) in cancer development and progression.

"PKM2 is very active during infancy, when you want rapid cell growth, and eventually it turns off. Tumor cells turn PKM2 back on - it's overexpressed in many types of cancer," said Zhimin Lu, M.D., Ph.D., the paper's senior author and an associate professor in MD Anderson's Department of Neuro-Oncology.

Lu and colleagues showed earlier this year that PKM2 in the nucleus also activates a variety of genes involved in cell division. The latest paper shows how it triggers aerobic glycolysis, processing glucose into energy, also known as the Warburg effect, upon which many types of solid tumors rely to survive and grow.

"PKM2 must get to the nucleus to activate genes involved in cell proliferation and the Warburg effect," Lu said. "If we can keep it out of the nucleus, we can block both of those cancer-promoting pathways. PKM2 could be an Achilles' heel for cancer."

By pinpointing the complicated steps necessary for PKM2 to penetrate the nucleus, Lu and colleagues found a potentially druggable target that could keep the protein locked in the cell's cytoplasm.

MEK, ERK emerge as targets

The process begins when the epidermal growth factor connects to its receptor on the cell surface. This leads to:

• Activation of the MEK protein, which in turn activates ERK.

• ERK sticking a phosphate group to a specific spot on PKM2.

• Phosphorylation priming PKM2 for a series of steps that culminate in its binding to the protein importin, which lives up to its name by taking PKM2 through the nuclear membrane.

Once in the nucleus, the team showed that PKM2 activates two genes crucial to aerobic glycolysis and another that splices PKM RNA to make even more PKM2.

An experiment applying several kinase-inhibiting drugs to human glioblastoma cell lines showed that only a MEK/ERK inhibitor prevented EGF-induced smuggling of PKM2 into the nucleus. ERK activation then is mandatory for PKM2 to get into the nucleus.

"MEK/ERK inhibitors have not been tried yet in glioblastoma multiforme," Lu said. Phosporylated PKM2 is a potential biomarker to identify patients who are candidates for MEK/ERK inhibitors once those drugs are developed.

MEK inhibitor blocks tumor growth

The researchers also found that the two glycolysis genes activated by PKM2, called GLUT1 and LDHA, are required for glucose consumption and conversion of pyruvate to lactate, crucial factors in the Warburg Effect. Depleting PKM2 in tumor cell lines reduced glucose consumption and lactate production.

In mice, depleting PKM2 blocked the growth of brain tumors. Re-expressing the wild type protein caused tumors to grow. However, re-expression of a PKM2 mutant protein that lost its ability to get into the nucleus failed to promote tumor formation. Experiments in human glioblastoma cell lines showed the same effect.

Injecting the MEK inhibitor selumetinib into tumors inhibited tumor growth, reduced ERK phosphorylation, PKM2 expression and lactate production in mice. In 48 human tumor samples, the team found that activity of EGFR, ERK1/2 and PKM2 were strongly correlated.

Cause of PKM2 overexpression

Lu and colleagues also published a paper in Molecular Cell that revealed a mechanism for overexpression of PKM2 in glioblastoma. They found that EGF receptor activation turns on NF-?B, which leads to a series of events culminating in PKM2 gene activation.

PKM2 levels were measured in tumor samples from 55 glioblastoma patients treated with standard of care surgery, radiation and chemotherapy. The 20 with low PKM2 expression had a median survival of 34.5 months, compared to 13.6 months for the 35 patients with high levels of PKM2.

Level of PKM2 expression in 27 low-grade astrocytomas was about half of the expression found in higher grade glioblastomas.

"In these two papers, we show how PKM2 is overexpressed in tumors, how it gets into the nucleus, that nuclear entry is essential to tumor development, and identified potential drugs and a biomarker that could usefully treat people," Lu said.

Arafat's remains exhumed for poison tests

RAMALLAH, West Bank (Reuters) - Forensic experts took samples from Yasser Arafat's buried corpse in the West Bank on Tuesday, trying to determine if he was murdered by Israeli agents using the hard-to-trace radioactive poison, Polonium.

Palestinians witnessed the funeral of their hero and longtime leader eight years ago, but conspiracy theories surrounding his death have never been laid to rest.

Many are convinced their icon was the victim of a cowardly assassination, and may stay convinced whatever the outcome of this autopsy. But some in the city of Ramallah where he lies deplored the exhumation.

"This is wrong. After all this time, today they suddenly want to find out the truth?" said construction worker Ahmad Yousef, 31, who stopped to watch the disinterment, carried out behind a wall of blue plastic near the Palestinian presidency headquarters.

"They should have done it eight years ago," he said.

French magistrates in August opened a murder inquiry into Arafat's death in Paris in 2004 after a Swiss institute said it had discovered high levels of polonium on clothing of his which was supplied by his widow, Suha, for a television documentary.

"Samples will be taken according to a very strict protocol and these samples will be analyzed," said Darcy Christen, spokesman for Lausanne University Hospital in Switzerland that carried out the original tests on Arafat's clothes.

"In order to do these analyses, to check, cross-check and double cross-check, it will take several months and I don't think we'll have anything tangible available before March or April next year," he added.

Arafat was always a freedom fighter to Palestinians but a terrorist to Israelis first, and a partner for peace only later. He led the bid for a Palestinian state through years of war and peacemaking, then died in a French hospital aged 75 after a short, mysterious illness.

No autopsy was carried out at the time, at the request of Suha, and French doctors who treated him said they were unable to determine the cause of death.

But allegations of foul play immediately surfaced, and many Palestinians pointed the finger at Israel, which confined Arafat to his West Bank headquarters in Ramallah for the final two and a half years of his life after a Palestinian uprising erupted.

Israel denies murdering him. Its leader at the time, Ariel Sharon, now lies in a coma from which he is expected never to awake. Israel invited the Palestinian leadership to release all Arafat's medical records, which were never made public following his death and still have not been opened.

FRENCH INVESTIGATORS

Polonium, apparently ingested with food, was found to have caused the death of former Russian spy Alexander Litvinenko in London in 2006. But some experts have questioned whether Arafat could have died in this way, pointing to a brief recovery during his illness that they said was not consistent with radioactive poisoning. They also noted he did not lose all his hair.

Eight years is considered the limit to detect any traces of the fast-decaying polonium and Lausanne hospital questioned in August if it would be worth seeking any samples, if access to Arafat's body was delayed as late as "October or November."

Not all of Arafat's family agreed to the exhumation, and his wife Suha chose not to attend the operation she had prompted.

Working in parallel with the forensic team, French magistrates were in Ramallah this week to ask if members of Arafat's inner circle might be able to shed light on his death.

One source told Reuters the French had a list of 60 questions, and had questioned one man for five hours.

Many Palestinians acknowledge that a Palestinian would almost certainly have had to administer any poison, wittingly or unwittingly.

College students more eager for marriage than their parents are

ScienceDaily (Nov. 28, 2012) ? Reaching adulthood certainly takes longer than it did a generation ago, but new research shows one way that parents are contributing to the delay.

A national study found that college students think 25 years old is the "right age" to get married, while a majority of parents feel 25 is still a little too soon. So it's no coincidence that when Justin Bieber said he'd like to wed by 25, Oprah Winfrey urged him to wait longer.

"The assumption has been that the younger generation wants to delay marriage and parents are hassling them about when they would get married," said Brian Willoughby, a professor at Brigham Young University and lead author of the study. "We actually found the opposite, that the parental generation is showing the 'slow down' mindset more than the young adults."

Willoughby and his co-authors in BYU's School of Family Life gathered info from 536 college students and their parents from five college campuses around the country (BYU was not in the sample). As they report in The Journal of Social and Personal Relationships,the scholars found the hesitation is consistent across gender.

"Initially we thought that this might be dads wanting their daughters to delay marriage," Willoughby said. "Moms and dads trended together -- gender wasn't a factor."

One of the driving forces behind parents' restraint is the feeling that their children should get an education first. While they generally feel marriage is important, parents think the "right age" is one year older than what their children say. Excluding teen marriages, research doesn't support the notion that there is an optimal time to tie the knot.

"I think parents have a lot of fear for their kids that makes them want to delay the transitions to adulthood," Willoughby said.

According to Census data, the median age for first marriages is 27. Willoughby says that what people say is the "right age" generally comes a few years before the actual marriage age.

"What happens is that someone thinks that 25 is when they want to get married," Willoughby said. "So at age 25, they start changing their patterns around dating, and it takes two or so years to make the transition."

Though BYU students weren't in Willoughby's sample, the university's own records show about 25 percent of its students are married. Willoughby said that Mormon young adults typically marry about two years younger than their peers nationally and have risen in sync with national trends.

Chad Olsen, a graduate student in BYU's School of Family Life, is a co-author on the new study. Professors Jason Carroll, Larry Nelson and Rick Miller are also co-authors.

A freight train slammed into a parade float carrying wounded veterans on Thursday, killing four people and injuring 17 others as the float drove through a West Texas railroad crossing on its way to an honorary banquet, authorities said.

The eastbound train was sounding its horn before it hit the float around 4:40 p.m. in Midland, Union Pacific spokesman Tom Lange said. A preliminary investigation indicates the crossing gate and lights were working at the time, Lange said, though he didn't know if the train crew saw the float approaching.

Two people died at the scene of the crash, while two others died at Midland Memorial Hospital, City of Midland spokesman Ryan Stout said. Seven of those injured are in critical condition, while the 10 others are in stable condition, he said.

"There is going to be a very thorough investigation," Lange said. "It's obviously a very tragic incident."

Photos of the float taken during the parade show about two dozen people seated in chairs set up on the back of a flatbed tractor-trailer decorated with American flags and signs identifying each veteran. A banner across the truck's front bumper reads, "Heroes on Board."

The float was among two flat-bed trucks carrying veterans and their spouses, police said. The first truck safely crossed the railroad tracks, but the second truck's trailer was hit by the train. Police said some of the people on the second trailer were able to evacuate before the crash.

The parade was to end at a "Hunt for Heroes" banquet honoring the veterans. The wounded service members were then going to be treated to a deer-hunting trip this weekend. The events have been canceled.

Lange said Union Pacific is offering help to the community and victims' families, as well as peer-to-peer counseling for the train crew, who did not sustain any injuries.

"It's pretty traumatic for them," he said.

The National Transportation Safety Board also is investigating, NTSB spokesman Peter Knudson said.

Midland is about 320 miles west of Dallas.

The musical drama "A Late Quartet" is a lot more compelling when it discusses music than when it focuses on the drama.

A string quartet is marking its 25th anniversary, but there?s turmoil among the long-harmonizing foursome. The group?s cellist and senior member, Peter (Christopher Walken), has been diagnosed with Parkinson?s disease and wants the other three to start planning for his replacement. The second violinist, Robert (Philip Seymour Hoffman), thinks Peter?s departure opens a door for him to occasionally take the first-violin duties. Daniel (Mark Ivanir), the perfectionist first violinist, thinks this will destroy the quartet?s sound ? and it?s a blow to Robert?s ego when the violist, Robert?s wife, Juliette (Catherine Keener), agrees with Daniel. Oh, and by the way, Daniel starts a relationship with Robert and Juliette?s daughter Alexandra (Imogen Poots), an up-and-coming violinist.

Opens Friday, Nov. 16, at the Broadway Centre Cinemas; rated R for language and some sexuality; 105 minutes.

The script ? by director Yaron Silberman and co-writer Seth Grossman ? is fascinating, even poetic, when it discusses the joys of music and connects the harmony of the quartets (specifically, Beethoven?s Op. 131) with the harmony of the musicians playing it. But the melodrama among the main characters is more soap opera than grand opera, even with a quartet of actors as fine as this.

Obama wants 'conversation' on climate change

WASHINGTON (AP) ? President Barack Obama said Wednesday that his administration has not done enough to combat global warming but said he hopes to begin his second term by opening a national "conversation" on climate change.

Obama said at a news conference that he took some steps in his first term to slow global warming, such as sharply increasing fuel efficiency standards for cars and trucks.

"But we haven't done as much as we need to," Obama said in his second comments on global warming since winning re-election last week.

Climate change was virtually absent during the presidential campaign until Hurricane Sandy hit the East Coast. The devastating superstorm ? a rarity for the Northeast ? and an election that led to Democratic gains have elevated global warming as a subject of renewed political debate.

Obama said during his victory speech in Chicago last week that Americans "want our children to live in an America that isn't ... threatened by the destructive power of a warming planet."

On Wednesday, Obama did not outline specific legislation, but said he would talk with scientists, engineers and elected officials to find ways to make short-term progress to reduce carbon emissions.

After that, he said the country should begin long-term efforts "to make sure that this is not something we're passing on to future generations," noting that floods, hurricanes and other disasters exacerbated by climate change are "going to be very expensive and very painful to deal with."

Obama did not mention a possible carbon tax pushed by some activist groups. A White House official said this week no such proposal is on the table.

Taking on climate change in a serious way will require "tough political choices" at a time when Americans are more focused on the economy and jobs, Obama said. "If the message is somehow we're going to ignore jobs and growth simply to address climate change, I don't think anybody's going to go for that. I won't go for that."

But if Republicans and Democrats can shape an agenda that helps create jobs and makes "a serious dent in climate change," then the American people will be supportive, Obama said.

The right-leaning American Enterprise Institute held an all-day discussion Tuesday on a possible carbon tax, which would make people pay more for using fossil fuels such as coal, oil, and gas that produce heat-trapping carbon dioxide and other greenhouse gases. The more liberal Brookings Institution has released a "modest" carbon tax proposal that would raise $150 billion a year, with $30 billion annually earmarked for clean energy investments.

Brad Johnson, campaign manager for ClimateSilence.org, an environmental group, said he welcome Obama's "belated call for a national conversation about how to address climate pollution."

But Johnson said Obama's assertion that climate change should be secondary to economic concerns was "a gross disappointment and an insult to the deep suffering of the millions of victims of climate disasters across this nation," including Hurricane Sandy. Obama is scheduled to tour New York City Thursday to view storm damage and recovery efforts.

"While conventional D.C. wisdom is focused on the manufactured crisis of the 'fiscal cliff,'" Johnson said, "the truth is that the most urgent threat to our national safety and economic well-being is the climate cliff that we are already beginning to tumble over."

Police: Deaths of 5 in Toledo, Ohio garage do not appear accidental

Five people were found dead of apparently carbon monoxide poisoning in a garage at a Toledo, Ohio, home on Monday afternoon, and police said the deaths did not appear accidental.

The victims were a 56-year-old woman, her three grandchildren ? ages 10, 7 and 5 ? and a 32-year-old man who was the uncle of one of the grandchildren,?according to the Toledo Blade newspaper.

Firefighters found the dead at 3:36 p.m. after being called by a family member to conduct a safety check. They used equipment to break into the garage.

Authorities did not say whether a carbon monoxide detector was in the home, NBC 24 in Toledo reported.

The problem: a glut of cheap Chinese solar panels that has forced many US and European panelmakers out of business. Even Chinese companies appear uncertain of survival, because competition has driven prices below break-even. Ironically, the cheap panels have spurred a boom in solar installations at the very time that solar manufacturers are losing their shirts.

The glut has also strained trade relations. The United States and European Union have imposed tariffs or are looking to do so. The Chinese are retaliating by investigating polysilicon it imports from the US and EU to make its solar panels.

There are still ways the players can avoid an all-out trade war.?But the tit-for-tat escalations are not helping.?

A year ago, seven US solar manufacturers complained to the US International Trade Commission that Chinese solar companies were getting Chinese government subsidies and were dumping their products on the US market. A month later, China initiated its own investigation into US solar subsidies. Undeterred, the US imposed?tariffs?earlier this year and, on Wednesday, affirmed that duties of some 24 to 36 percent on most Chinese solar panels was proper.

An even bigger trade fight is brewing in Europe, which represents nearly 75 percent of world demand for solar panels. In September, the European Union said it would investigate possible dumping of Chinese solar products. On Nov. 1, China fired back, saying it would investigate solar-grade polysilicon imports from the EU.

On Thursday, the European Union said it is also considering?tariffs?on Chinese-made?solar?panels and parts because of Chinese subsidies.

There's still room for compromise. For one thing, Chinese manufacturers are already changing where they manufacture the panels, reports suggest, in order to get around US tariffs. The supply-chain maneuvers may raise costs by up to 15 percent, according to one estimate, but the glut is so big that it's expected to have little if any effect on prices.

For another thing, the US solar industry is hardly united behind tariffs against the Chinese. While panel manufacturers have felt the squeeze and support the anti-Chinese move, installers oppose it because they are benefiting handsomely from cheap panels. A September report from?GTM Research and the Solar Energy Industries Association forecast that a total 3.2 gigawatts of photovoltaic panels will be installed in the US in 2012, a 71 percent increase over 2011.

Indeed, the solar-panel manufacturer leading the charge for tariffs in the US is owned by a German company, SolarWorld, which has been leading the EU industry's push for tariffs as well.?

For their part, the Chinese are unlikely to push so hard as to alienate a major trading partner such as the US. It certainly cannot afford to offend its even bigger trading partner, the EU. Moreover, Germany is gradually reducing its subsidies to the solar industry.

Despite this reduction in government aid, solar power production in Germany jumped by more than half this year over the first nine months of 2011, according to BDEW, the German utilities' industry association.?

All three regions ? the US, the EU, and China ? have targeted solar manufacturing as an industry of the future that will generate new jobs. But their policies have created a glut that threatens the survival of many of those manufacturers.?

They may have to rethink their strategies.?

SOS Crock-Pot Chili is my recipe that along with many other food bloggers is to support the victims of Hurricane Sandy. We were asked to make a dish that would be what we would feed to those whose lives were forever changed by Sandy. Living in Texas, chili comes to mind. Living through a few hurricanes myself I know what a good home-cooked meal can do to help raise the spirits.


SOS Crock-Pot Chili is a very easy chili to put together, it has very common ingredients, and by using a Crock-Pot we have a meal that almost cooks itself.
It is my hope that you consider a donation to a charitable organization to assist those whose lives were impacted by this tragedy.

SOS Crock-Pot Chili

SOS Crock-Pot Chili is a hearty chili that can be made in a Crock-Pot, this recipe features smoked paprika, and chipotle powder to give this recipe a little something extra.

Ingredients

• 2 pounds ground beef
• 1 cup chopped onion
• 1 tablespoon chopped garlic
• 1/2 cup chopped celery
• 2 15 ounce cans chopped tomato
• 1 15 ounce can tomato sauce
• 1 15 ounce can pinto beans
• 1 15 ounce can red kidney beans
• 4 tablespoons chili powder
• 1 tablespoon cumin
• 1 teaspoon smoked paprika
• 1 teaspoon chipotle powder
• additional liquid if desired you can use tomato juice, beer, or water

Instructions

In a skillet over medium high heat cook together the ground beef, onion, chopped garlic, and celery. Cook until the ground beef is no longer pink. Using a 6 quart Crock-Pot add chopped tomatoes, tomato sauce, pinto beans, red kidney beans, and spices. When ground beef has finished cooking drain off grease and add to the crock pot. Stir well. If you desire more liquid in your chili you can add tomato sauce, or beer, or water until it has reached the consistency you desire. Cook on low for 6 to 8 hours.

Criminally charged in September with stealing from clients, disbarred last month and ordered in a separate civil eviction case to vacate by Oct. 31 the Dallas law office for which he had failed to pay rent, a former Texas personal injury lawyer is now facing new trouble.

Thomas Corea, who once hosted a live attorney call-in television show in better days, was jailed Wednesday morning after allegedly trashing his Design District law office premises prior to his eviction and causing as much as $100,000 in damage, the Crime Blog of the Dallas Morning News reports.

State District Judge Mike Snipes increased Corea's bail this morning to $500,000 on each of four felony charges in the Dallas County theft and client identity fraud case he faces, after a prosecutor asked the jurist to find that Corea's bond was insufficient because of the damage to the law office, the newspaper reports..

The Dallas Observer's Unfair Park blog includes a number of photos of the claimed damage, including graffiti. The scribbling on the law office walls reportedly included a derogatory reference to an Arizona judge who had found Corea in contempt in an unrelated case, along with her apparent phone number, which is in the 505 area code, and a suggestion that readers of the graffiti call her.

"I doubt seriously there are any random vagrants in the Dallas area that are familiar with area code 505,? Snipes said Wednesday as he made the bond ruling that sent Corea to jail, at least for now. The judge told Corea's lawyer, John Gussio, that he may reconsider if more information is provided later, once the defense lawyer has a chance to prepare further for a hearing, Crime Blog reported.
